Effect of dietary fish oil on serum lipids and lipoproteins of rats fed a hyperlipidemic diet

Abstract
This investigation was attempted in order to clarify the effects of fish oil on serum lipids and lipoproteins in rats fed a hyperlipidemic diet. Male Sprague-Dawley rats were maintained on a semipurified high fat diet supplemented with 7.3 % coconut oil, 7.3 % beef tallow, 0.4 % safflower oil and 1.5 % cholesterol in order to induce hyperlipidemia within 28 days. These rats were than switched to a fish oil diet with 1.4 %, 2.8 % and 5.6 % fish on for respective 10 and 20 days.Feeding a hyperlipidemic diet increased the levels of serum cholesterol by about 26 %. This was mainly due to a 5-fold increase in VLDL cholesterol, wheras also HDL cholesterol raised. A hyperlipidemic diet did not significantly effect serum, LDL and HDL concentration of triglycerides. VLDL triglycerides increased only after 28 and 48 days. Also a hyperlipidemic diet did not markedly alter serum concentration of phospholipids, but caused an almost 2-fold increase in VLDL phospholipids. In all rats on fish oil diets, there was depression of serum and VLDL concentration of cholesterol. Serum triglycerides tended to be lower in rats fed fish oil. Especially after 20 days VLDL triglycerides significantly fell. However, there was no consistent effect on LDL and HDL cholesterol and triglycerides. Substitution of fish oil for coconut oil and beef tallow especially lowered the phospholipid concentration in serum and lipoproteins. LDL phospholipids were reduced only after 20 days on fish oil diet given in higher doses. These findings elucidate that fish oil especially lowers serum cholesterol and phospholipids as well as total VLDL in rats with hypercholesterolemia experimentally induced.
